Summary for clan CF

Summary Alignment Structure Literature
Clan type peptidaseC15.001 - pyroglutamyl-peptidase I (prokaryote) (Thermococcus litoralis), MEROPS Accession MER0003453 (peptidase unit: 1-220); PDB accession 1A2Z_A
HistoryMEROPS 3.03 (9 September 1998)
DescriptionCysteine nucleophile; catalytic residues in the order Glu, Cys, His in sequence
Contents of clanThe clan contains a single family, C15.
EvidenceThe structure of pyroglutamyl-peptidase I shows a protein fold unlike that of any other cysteine peptidase, so pyroglutamyl-peptidase I provides the type structure for clan CF. There are structural similarities detectable by the Dali program (Holm & Sander, 1997) between members of clan CF and three clans of metallopeptidases, MC, MF and MH, and all four peptidase clans are included in the phosphorylase/hydrolase-like fold by the SCOP database.
Protein foldThe tertiary structure for pyroglutamyl-peptidase I has been determined (Odagaki et al., 1999) and shows an alpha/beta protein with an alpha/beta/alpha sandwich. The beta sheet contains five strands (in the order 21354) with strand 4 antiparallel to the others.
