Over 300 PLP enzymes have been identified up to the year 2000 [1].

PLP-dependant enzymes can be classified into 4 groups, based on protein fold [1]:

  1. α- and γ-families (si-specific)
  2. β-family (si-specific)
  3. both specificities
  4. re-specific

7,8-Diaminopelargonic acid synthase uses PLP and SAM as cofactor. In the E. coli enzyme PLP is covalently bound to a lysine residue. SAM uniquely acts as an amino donor. This enzyme is the second step in biotin biosynthesis [3].
