DefinitionTPT-172 (R33) is a thiophene-thiourea derivative with a molecular weight of 172 (free base) that exhibits potent preclinical activity by stabilizing the proper folding and trafficking of misfolded proteins. In vitro studies demonstrate that TPT-172 enhances cellular processing of target proteins, improving their localization and functional activity while reducing accumulation of misfolded or aggregated species. In cellular models, it restores normal protein homeostasis and reduces endoplasmic reticulum stress.
