Within the pathway, mfmB catalyzes the first step that leads to the synthesis of 3,5-dimethylorsellinic acid (DMOA) via condensation of one acetyl-CoA starter unit with 3 malonyl-CoA units and 2 methylations (PubMed:38404388). DMOA is then transformed into the phthalide 5,7-dihydroxy-4-(hydroxymethyl)-6-methylphthalide by the cytochrome P450 monooxygenase mfmA and the hydrolase mfmC. Subsequently, the methyltransferase mfmE catalyzes 7-O-methylation to yield 5-hydroxy-4-(hydroxymethyl)-7-methoxy-6-methylphthalide, which undergoes C-3 hydroxylation by the cytochrome P450 monooxygenase mfmF. The resultant cyclopolic acid (2,5-dihydroxy-4-(hydroxymethyl)-7-methoxy-6-methylphthalide) is then farnesylated by the DMATS-type prenyltransferase mfmD to afford 5-O-farnesylcyclopolic acid. Finally, the Pyr4-family terpene cyclase mfmH cyclizes the farnesyl moiety of 5-O-farnesylcyclopolic acid into a drimane-like structure, thus completing the biosynthesis of 11'-O-desmethylfendlerol (PubMed:38404388)"
