Webinar
Using in situ cryoET to investigate intracellular host-pathogen interactions
This webinar will focus on methods of in situ cryo electron tomography (cryoET) and how they can be applied to specifically look at host-pathogen interactions inside the host cell with a focus on apicomplexan parasites and gram-negative bacteria. From sample preparation to data acquisition and data analysis, this webinar will cover approaches of sample freezing, cryo focussed ion beam (cryoFIB) milling, cryoET, cryo correlative light and electron microscopy (cryoCLEM), tomogram reconstruction, segmentation and subtomogram averaging.
This event is part of the webinar series “Integrating structural biology and bioinformatics to study infection”. You can follow the link for more information about the series and its webinars.
Who is this course for?
Students and researchers that are interested to know what aspects of the host-pathogen interface can be investigated using in situ cryoET. Coarse understanding of the biology of apicomplexan parasites/gram-negative bacteria and cryo electron microscopy is advantageous but not required.
Outcomes
By the end of the webinar, you will be able to:
Have an overview of the in situ cryoET workflow and its different approaches
Describe the advantages and limitations of in situ cryoET
Identify when in situ cryoET is suitable for your research question
