| UniProt code: Q6IVU0. |
| Organism: | Machupo virus (MACV). | |
| Taxonomy: | Viruses; Riboviria; Orthornavirae; Negarnaviricota; Polyploviricotina; Ellioviricetes; Bunyavirales; Arenaviridae; Mammarenavirus. | |
| Function: | RNA-dependent RNA polymerase which is responsible for replication and transcription of the viral RNA genome. During transcription, synthesizes 4 subgenomic RNAs, and assures their capping by a cap-snatching mechanism, in which cellular capped pre-mRNAs are used to generate primers for viral transcription. The 3'-end of subgenomic mRNAs molecules are heterogeneous and not polyadenylated. The replicase function is to direct synthesis of antigenomic and genomic RNA which are encapsidated and non capped. As a consequence of the use of the same enzyme for both transcription and replication, these mechanisms need to be well coordinated. These processes may be regulated by proteins N and Z in a dose-dependent manner. {ECO:0000256|HAMAP-Rule:MF_04086, ECO:0000256|PIRNR:PIRNR000836}. | |
| Catalytic activity: | Reaction=a ribonucleoside 5'-triphosphate + RNA(n) = diphosphate + RNA(n+1); Xref=Rhea:RHEA:21248, Rhea:RHEA-COMP:11128, Rhea:RHEA- COMP:11129, ChEBI:CHEBI:33019, ChEBI:CHEBI:61557, ChEBI:CHEBI:83400; EC=2.7.7.48; Evidence={ECO:0000256|HAMAP-Rule:MF_04086, ECO:0000256|PIRNR:PIRNR000836}; | |
| Subunit: | Homomultimerizes; the oligomeric structure is essential for the polymerase activity. Interacts with the nucleocapsid protein N. Interacts with protein Z; this interaction inhibits viral transcription and replication. {ECO:0000256|HAMAP-Rule:MF_04086, ECO:0000256|PIRNR:PIRNR000836}. | |
| Subcellular location: | Virion {ECO:0000256|HAMAP-Rule:MF_04086, ECO:0000256|PIRNR:PIRNR000836}. Host cytoplasm {ECO:0000256|HAMAP- Rule:MF_04086, ECO:0000256|PIRNR:PIRNR000836}. | |
| Domain: | The N-terminal domain contains an RNA endonuclease activity that plays an essential role in mRNA transcription. This activity may be involved in cap snatching. {ECO:0000256|HAMAP-Rule:MF_04086}. | |
| Similarity: | Belongs to the arenaviridae RNA polymerase family. {ECO:0000256|HAMAP-Rule:MF_04086, ECO:0000256|PIRNR:PIRNR000836}. | |
