Small-molecule inhibitor: danoprevir

Summary Structure Literature

Name

Common name
danoprevir
Other names
ITMN-191; R7227; RG7227

Inhibition

Peptidases inhibited
Inhibits hepacivirin.
Mechanism
A study of the mechanism of action of danoprevir provided results consistent with the formation of an initial collision complex between the enzyme and the inhibitor that isomerizes to a highly stable complex that dissociates only very slowly, so that the effective Ki is sub-nanomolar (Rajagopalan et al., 2009).
Pharmaceutical relevance
Danoprevir shows promise for the treatment of hepatitis C (Forestier et al., 2011).
