Summary for family M88

Family type peptidaseM88.001 - IMPa peptidase (Pseudomonas aeruginosa) (Pseudomonas aeruginosa), MEROPS Accession MER0185642 (peptidase unit: 181-923)
Content of familyFamily M88 contains endopeptidases.
History Identifier created: MEROPS 9.7 (1 August 2012)
The human pathogenic bacterium Pseudomonas aeruginosa evades immunological attack by secreting peptidases that degrade components of the host defence system, such as pseudolysin (M04.005) and aeruginolysin (M10.056). IMPa ('immunomodulating protease'; M88.001) is another example of a secreted Pseudomonas peptidase that interferes with the immune system by degrading PSGL-1 (P-selectin glycoprotein ligand 1), which by interacting with selectins mediates the migration of leukocytes over vascular surfaces ('rolling') during the initial stages of inflammation (Bardoel et al., 2012).
Catalytic typeMetallo
Active site residuesH696 E697 H700
Active siteIMPa is assumed to be a metallopeptidase because of the presence of a well-conserved metal-binding motif, HEXXH, in which the histidines are assumed to be two of the three zinc ligands and the glutamic acid a catalytic residue. The third zinc ligand has not been identified and cannot be predicted from the alignment.

Biological functions IMPa protects the bacterium from immunological attack by impairing the extravasation of neutrophils (movement of the cells from the capillaries to the surrounding tissue; Bardoel et al., 2012%).
