Subsequently, the methyltransferase mfmE catalyzes 7-O-methylation to yield 5-hydroxy-4-(hydroxymethyl)-7-methoxy-6-methylphthalide, which undergoes C-3 hydroxylation by the cytochrome P450 monooxygenase mfmF. The resultant cyclopolic acid (2,5-dihydroxy-4-(hydroxymethyl)-7-methoxy-6-methylphthalide) is then farnesylated by the DMATS-type prenyltransferase mfmD to afford 5-O-farnesylcyclopolic acid.
