Activated by amino acids through a mechanism involving the lysosomal V-ATPase, the Ragulator plays a dual role for the small GTPases Rag (RagA/RRAGA, RagB/RRAGB, RagC/RRAGC and/or RagD/RRAGD): it (1) acts as a guanine nucleotide exchange factor (GEF), activating the small GTPases Rag and (2) mediates recruitment of Rag GTPases to the lysosome membrane. Activated Ragulator and Rag GTPases function as a scaffold recruiting mTORC1 to lysosomes where it is in turn activated. When complexed to BIRC5, interferes with apoptosome assembly, preventing recruitment of pro-caspase-9 to oligomerized APAF1, thereby selectively suppressing apoptosis initiated via the mitochondrial/cytochrome c pathway"],"length":90,"sequence":"MSLSFHLPSRRMKNPSIVGVLCTDSQGLNLGCRGTLSDEHAGVISVLAQQAAKLTSDPTDIPVVCLESDNGNIMIQKHDGITVAVHKMAS","proteome":"UP000233220","gene":"LAMTOR5","go_terms":[{"identifier":"GO:0043066","name":"negative regulation of apoptotic process","category":{"code":"P","name":"biological_process"}},{"identifier":"GO:0005737","name":"cytoplasm","category":{"code":"C","name":"cellular_component"}},{"identifier":"GO:0071986","name":"Ragulator complex","category":{"code":"C","name":"cellular_component"}}],"protein_evidence":3,"source_database":"unreviewed","is_fragment":false,"in_alphafold":true,"in_bfvd":false,"ida_accession":"f229e81e6b48a12154f968815ad9df41fb51441f","counters":{"domain_architectures":1739,"entries":5,"isoforms":0,"proteomes":1,"sets":1,"structures":0,"taxa":1,"dbEntries":{"cathgene3d":1,"pfam":1,"panther":1,"prints":1,"interpro":1},"proteome":1,"taxonomy":1,"similar_proteins":1739}}}
