The 'bivalent Mical/EHBP Rab binding' (bMERB) domain is a Rab effector domain that is present in proteins of the Mical and EHBP families, both known to act in endosomal trafficking. The bMERB domain displays a preference for Rab8 family proteins (Rab8, 10, 13 and 15) and at least some of the bMERB domains contain two separate binding sites for Rab-proteins, allowing Micals and EHBPs to bind two Rabs simultaneously. The strong similarity between the two binding sites within one bMRB domain strongly suggests an evolutionarily development via duplication of a common ancestor supersecondary structure element. The bMERB domain has a completely alpha-helical fold consisting of a central helix and N- and C-terminal helices folding back on this central helix [[cite:PUB00088663]].</p>","llm":false,"checked":false,"updated":false}],"wikipedia":null,"literature":{"PUB00088663":{"PMID":27552051,"ISBN":null,"volume":"5","issue":null,"year":2016,"title":"bMERB domains are bivalent Rab8 family effectors evolved by gene duplication.","URL":null,"raw_pages":null,"medline_journal":"Elife","ISO_journal":"Elife","authors":["Rai A","Oprisko A","Campos J","Fu Y","Friese T","Itzen A","Goody RS","Gazdag EM","Muller MP."],"DOI_URL":"https://doi.org/10.7554/eLife.18675"}},"set_info":null,"overlaps_with":null,"counters":{"subfamilies":0,"domain_architectures":118,"interactions":0,"matches":13023,"pathways":0,"proteins":12866,"proteomes":1275,"sets":0,"structural_models":{"alphafold":7449},"structures":14,"taxa":4099},"entry_annotations":{"hmm":0,"logo":0,"alignment:seed":57,"alignment:full":9721},"cross_references":{},"is_llm":false,"is_reviewed_llm":false,"is_updated_llm":false,"representative_structure":{"accession":"6zsi","name":"The mechanism of activation of the actin binding protein EHBP1 by Rab8 family members."}}}
