Resolution of HJs is mediated by a diverse group of DNA structure specific endonucleases known as Holliday junction resolvases (HJR) [[cite:PUB00108910]]. This entry is specific for RuvX also known as YqgF a family of nucleases which resolves the Holliday junction intermediates in genetic recombination[[cite:PUB00009701],[cite:PUB00001215]]. Studies carried out in M. tuberculosis, have shown that YqgF/RuvX is a genuine HJR analogous to RuvC from E. coli. Furthermore, a single cysteine present in M. tuberculosis RuvX was found to be required for disulfide-bond mediated intermolecular dimerization and HJ resolution activity, suggesting that M. tuberculosis RuvX has adapted its YqgF protein to function like a typical RuvC family HJR [[cite:PUB00108910]].</p>","llm":false,"checked":false,"updated":false}],"wikipedia":null,"literature":{"PUB00108910":{"PMID":26817626,"ISBN":null,"volume":"100","issue":"4","year":2016,"title":"Mycobacterium tuberculosis RuvX is a Holliday junction resolvase formed by dimerisation of the monomeric YqgF nuclease domain.","URL":null,"raw_pages":"656-74","medline_journal":"Mol Microbiol","ISO_journal":"Mol Microbiol","authors":["Nautiyal A","Rani PS","Sharples GJ","Muniyappa K."],"DOI_URL":null},"PUB00009701":{"PMID":10982859,"ISBN":null,"volume":"28","issue":"18","year":2000,"title":"SURVEY AND SUMMARY: holliday junction resolvases and related nucleases: identification of new families, phyletic distribution and evolutionary trajectories.","URL":null,"raw_pages":"3417-32","medline_journal":"Nucleic Acids Res","ISO_journal":"Nucleic Acids Res.","authors":["Aravind L","Makarova KS","Koonin EV."],"DOI_URL":"http://dx.doi.org/10.1093/nar/28.18.3417"},"PUB00001215":{"PMID":1661673,"ISBN":null,"volume":"10","issue":"13","year":1991,"title":"Escherichia coli RuvC protein is an endonuclease that resolves the Holliday structure.","URL":null,"raw_pages":"4381-9","medline_journal":"EMBO J","ISO_journal":"EMBO J.","authors":["Iwasaki H","Takahagi M","Shiba T","Nakata A","Shinagawa H."],"DOI_URL":"http://ukpmc.ac.uk/picrender.cgi?tool=EBI&pubmedid=1661673&action=stream&blobtype=pdf"}},"set_info":{"accession":"CL0580","name":"YqgF"},"overlaps_with":null,"counters":{"subfamilies":0,"domain_architectures":56,"interactions":0,"matches":19371,"pathways":0,"proteins":19330,"proteomes":17943,"sets":1,"structural_models":{"alphafold":12884},"structures":7,"taxa":26077},"entry_annotations":{"hmm":0,"logo":0,"alignment:seed":98,"alignment:full":10056},"cross_references":{},"is_llm":false,"is_reviewed_llm":false,"is_updated_llm":false,"representative_structure":{"accession":"1nu0","name":"Structure of the double mutant (L6M; F134M, SeMet form) of yqgF from Escherichia coli, a hypothetical protein"}}}
