Chemically synthesized and HPLC-purified human chemokines, CXCL9/MIG were purchased from The Biomedical Research Centre, the University of the British Columbia (UBC), Vancouver, Canada. The chemotactic activities of these chemokines were checked by standard chemotaxis assays as described previously (Gong 1995). The chemokines were dissolved in endotoxin-free water (Sigma-Aidrich) and stored at -20 degree Celsius until further use. THP-1 cells (at density of 1 x 10^6 cells/ml) were treated with human chemokine MIG at a dose of 10 uM for 4 hours. The RNA was then isolated for microarray study. All experiments were done for at least three biological replicates with two technical replicates each.
