To generate polarized cell cultures, 16HBE14O- cells were seeded in six-well Transwell permeable plates (Corning Life science, New York, USA) at a density of 1.5 to 3.75 x 105 cells / well. Cells were grown in complete MEM plus penicillin-streptomycin (50 units/ml) (Invitrogen, Burlington, Canada) at 37oC in 5% CO2 until they were polarized (14 days). Polarization was monitored by measuring the trans-epithelial cell resistance with a Millicell electrical resistance system (Millipore, Billerica, USA). Medium was replaced every other day with fresh medium and the night before an experiment it was replaced with fresh complete MEM without antibiotics.
