The aortic segment between heart and diaphragm was dissected from the surrounding tissue, flushed with cold sterile phosphate-buffered saline (PBS), and cut open lengthwise. Approximately 10,000 endothelial cells were gently scraped from a region (~1 cm^2)located at the inner curve-lateral wall of the aortic arch (disturbed flow, DF), and separately from the dorsal descending thoracic aorta (undisturbed flow, UF). The cells were transferred directly to a lysis buffer containing the RNase inhibitors guanidine isothiocyanate and beta-Mercaptoethanol (0.143 M) (Qiagen Inc, Valencia, CA) and stored on dry ice.
