Insecticide resistance levels were measured in larvae and adults through bioassays with deltamethrin, DDT and bendiocarb. A microarray approach was used for identifying transcription level variations associated to different environments and insecticide resistance. the Ifakara strain originating from central Tanzania and susceptible to all insecticides was used as a reference strain. Version 6.6, September 2012
(Parameters: Scanning hardware = G2565AA DNA microarray scanner [Agilent], Scanning software = Feature Extraction Software [Agilent]) pools of twenty five 2-3 days-old adult females Anopheles arabiensis from each population were collected, stored in RNAlater and used for RNA extraction. Total RNA was extracted from pools of 25 three days-old adult females from each population using the RNAqueous-4PCR kit (Ambion)according to manufacturer's instructions. Total RNAs were treated with DNaseI (Ambion) to remove any genomic DNA contaminant.
(Parameters: Extracted product = total_RNA, Amplification = none) hybridization were performed according to the M-^QTwo-Color Microarray-Based Gene Expression Microarrays Analysis v.6.6' (G4140-900050).
(Parameters: Chamber type = Agilent, Quantity of label target used = 300, Mass unit = Nano gram, time = 17, Tiny time unit = hours, Volume unit = Nano litre, temperature = 65) Anopheles gambiae arabiensis mosquito larvae were collected from 9 different populations in east Tanzania (F0). Larvae were bred in insectary to the adult stage, allowed to mate and lay eggs for the next generation (F1). F1 individuals from each population were bred in standard insectary conditions and used for gene expression study using DNA microarray. Adult females of 2-3 days old (non-blood fed) were used for microarray experiments in comparison with individuals of same sex and life stage form the reference strain Ifakara.
