Protocol Description Total RNA was extracted from liver tissue by homogenation in Trizol.
RNA was purified using RNeasy mini kit protocol (Qiagen)
Fluorescently-labeled cRNA was generated using the Agilent Low RNA Input Fluorescent Linear Amplification Kit according tot the manufacturer's protocol.

The interclass correlation coefficient applied for evaluation of data correction, labeling methods, and rectal biopsy sampling in DNA microarray experiments. Physiol. Genomics 16, 99-106. RNA of all control animals was pooled Male fresian bull calves were purchased from the local market and housed for 2-3 weeks before the start of the experiment.Treatment with DHEA was repeated in three independent experiments using identical treatment schedules. Each of the three experiments consisted of two animals of which one was orally (PO) treated and the other was intramuscular (IM) treated. Administrations were performed seven times at 24 hour intervals. IM treated animals (n=3) were 9-12 months old, PO treated animals (n=3)were 8-13 months old. Control animals were included in the first (n=3, all 6 months old) and in the third (n=4, 13-14 months old)experiment. The animals were housed under normal livestock conditions. At the end of the experiment the animals were sacrificed, liver tissue was collected and snap frozen in liquid nitrogen.

The values were further log2 transformed. DHEA exposed animals wer treated either orally (PO) with capsules containing 1000 mg DHEA or intramuscular (IM) with 1000 mg DHEA disolved in 10 mL Miglyol. Administrations were performed 7 times at 24h intervals.
