Comment[ArrayExpressAccession] E-GEOD-35993 Public Release Date 2012-05-22 Investigation Title Notch regulated miRNAs in human T-ALL cell line Comment[Submitted Name] Notch regulated miRNAs in human T-ALL cell line Experiment Description In an attempt to identify miRNAs regulated by oncogenic Notch signaling, we performed miRNA profiling of human T-cell acute lymphoblastic leukemia (T-ALL) cells with or without the treatment of γ-secretase inhibitor (GSI) to block Notch signaling. We found miR-223 levels to increase after GSI treatment suggesting that active Notch signaling represses miR-223 expression. We confirmed insulin-like growth factor-1 receptor (IGF1R) to be regulated by miR-223, but were unable to demonstrate functional effects on T-ALL cell growth by overexpression or knock-down of miR-223 alone. 4 samples analyzed, 2 cell lines each treated (GSI) and mock treated Date of Experiment Term Source Name EFO Term Source Version Term Source File http://www.ebi.ac.uk/efo/efo.owl Person Last Name Weng Gusscott Weng Person First Name Andrew Sam Andrew Person Mid Initials P D P Person Email aweng@bccrc.ca Person Affiliation BC Cancer Agency Person Phone 604-675-8136 Person Fax 604-877-0712 Person Address Pathology, BC Cancer Agency, 675 West 10th Ave, Vancouver, BC, Canada Person Roles submitter Person Roles Term Source REF Person Roles Term Accession Number Normalization Type Normalization Term Accession Number Normalization Term Source REF Replicate Type Replicate Term Accession Number Replicate Term Source REF Experimental Design Experimental Design Term Accession Number Experimental Design Term Source REF Quality Control Type Quality Control Term Accession Number Quality Control Term Source REF Protocol Name P-GSE35993-1 P-GSE35993-6 P-GSE35993-3 P-GSE35993-8 P-GSE35993-7 P-GSE35993-2 P-GSE35993-4 P-GSE35993-5 Protocol Description ID_REF =

RPMI 1640 medium supplemented with 10% FCS, 1 mM sodium pyruvate, 2 mM L-glutamine, and antibiotics. Performed as per Exiqon standard procedures. Performed as per Exiqon standard procedures. Cells were treated with the gamma-secretase inhibitor (GSI) Compound E (E) at a concentration of 1uM for 4 days followed by washout and resuspension in media containing E or Mock (DMSO). Total RNA was extracted using standard Trizol protocol. Individual samples were labelled with Hy3 and compared to a common reference pool of all 4 sampled labelled with Hy5.
