Experimental samples are added and target proteins present in the samples bind to the immobilized antibodies. All wells also contain Reference Spots, which provide a strong positive signal to allow easy visualization of well location. A cocktail of biotinylated detection antibodies and Streptavidin-HRP or an HRP-conjugated pan anti-phospho-tyrosine antibody is subsequently used to detect the bound proteins. Chemiluminescent substrate reagents produce a signal that is proportional to the amount of analyte bound. A compatible digital imaging system capable of chemiluminescent detection is used to determine the intensity of light emitted from individual spots.
