Briefly, over night cultures of L. monocytogenes EGD-e cells (wild-type and sigma B mutant) were grown in brain heart infusion (BHI) broth (Difco). Bacteria were diluted 1:50 in 20 ml fresh BHI and incubated using a 100 ml Erlenmeyer flask at 37 °C with shaking (180 rpm, Unitron [Infors]). Bacterial cells were harvested at OD600 1.0 (3 h), OD600 2.0 (4 h), OD600 2.5 (8 h) OD600 2.5 (16 h) using RNAprotect (Qiagen) according to manufacturer.
