Literature Report

IntroductionN(10) Formyltetrahydrofolate synthetase (FTHFS) catalyses the ATP dependent activation of formate towards addition at the N10 position of tetrahydrofolate; a reaction of physiological importance to acetogens, catalysing the reduction of carbondioxide and other one carbon containing precursor molecules into acetate.
MechansimFirst, formate acts as a nucleophile towards the gamma phosphate of the Mg-ATP. This formyl phosphate intermediate then acts as an electrophile towards the N10 atom of Tetrahydrofolate (THF), resulting in the formation of 10-formyl-THF. No catalytic residues fitting the CSA definition of catalytic have been identified, although a number of residues involved in binding interactions with the triphosphate and tetrahydrofolate substrates have been determined. The presence of a metal cation enhances catalytic activity, but its binding site is too far from the active site to be implicated in the catalytic mechanism.
