TNF receptor-associated factor 6 (TRAF6) regulates a diverse array of processes, including adaptive immunity, innate immunity, bone metabolism and tissue development. It is a unique member of the TRAF family of adaptor proteins in that it is involved in both the TNF receptor superfamily and the interleukin-1 receptor (IL-1R)/Toll-like receptor (TLR) superfamily signal transduction pathways [PMID: 14579250, PMID: 17633022, PMID: 10215628, PMID: 14499111]. TRAF6 possesses ubiquitin ligase activity that controls the activation of IKK and NF-kappaB [PMID: 17135271, PMID: 15125833]. TRAF6 contains a RING finger domain, five zinc finger domains, and a TRAF domain.

The TRAF domain can be divided into a more divergent N-terminal alpha helical region (TRAF-N), and a highly conserved C-terminal MATH subdomain (TRAF-C) with an eight-stranded beta-sandwich structure. TRAF-N mediates trimerization while TRAF-C interacts with receptors [PMID: 10206649, PMID: 10518213].
