Praja-2 belongs to the mammalian RING E3-ubiquitin ligase family and has E2-dependent E3 ubiquitin-protein ligase activity. It regulates the total concentration of protein kinase A (PKA) regulatory (Rs) subunits, tuning the strength and duration of PKA signal output in response to cAMP [PMID: 21423175]. It binds to MOB1 (Mps one binder 1), a core effector of the Hippo pathway, and serves as an upstream regulator of the Hippo signalling, linking the ubiquitin proteasome system to deregulated glioblastoma growth [PMID: 23652010].
