Human CD81 is a transmembrane protein belonging to the tetraspanin family, whose members are characterised by four hydrophobic transmembrane regions (TM1-4) and two extracellular domains, known as large and small extracellular loops (LEL and SEL, respectively). This entry represents the LEL domain, which is topologically located between TM3 and 4 and contains four invariant Cys residues, two of which define a conserved Cys-Cys-Gly motif [PMID: 11226150]. The key structural features are conserved within the family.
