Four heat shock factor proteins have been identified in vertebrates (HSF1-4) [PMID: 10547058]. Heat shock factor protein 4 (HSF4) specifically binds heat shock promoter elements (HSE). HSF4 lacks the carboxyl-terminal hydrophobic domain (IPR010542) which is shared among all vertebrate HSFs. There are two alternatively spliced transcripts encoding distinct isoforms, HSF4A and HSF4B. HSF4A represses transcription while the isoform HSF4B activates transcription [PMID: 16371476]. HSF4B contains a PDSM phosphorylation-dependent sumoylation motif (PDSM) [PMID: 16371476].
