Virulence of pathogenic organisms such as the Gram-positive Streptococcus pneumoniae is largely determined by the ability to degrade host glycoproteins and to metabolise the resultant carbohydrates.

This domain represents the enzymatic region (EC: of cell surface glycoside hydrolases from family GH101 [PMID: 20556855] that specifically remove the T-antigen disaccharide (Gal-beta-1,3-GalNAc-alpha) linked to serine or threonine residues of mucin-type glycoproteins [PMID: 18784084]. This reaction is exemplified by the S. pneumoniae endo-alpha-N-acetylgalactosaminidase (SpGH101), where Asp764 is the catalytic nucleophile-base and Glu796 the catalytic proton donor [PMID: 19788271].
