Members of this family have been linked to DNA maintenance in bacteria and mitochondria [PMID: 19578062] and transcription regulation and telomere homeostasis in eukaryotes [PMID: 16874308, PMID: 16564010], but their function remained uncharacterised. A bacterial member of this family, Gcp, was originally thought to have endopeptidase activity [PMID: 1885539] but this could not be confirmed in other orthologues [PMID: 17766251, PMID: 21183954]. Recent research indicates that this family is involved in the biosynthesis of N6-threonylcarbamoyl adenosine, a universal modification found at position 37 of tRNAs that read codons beginning with adenine [PMID: 21285948].
