How fluorescence microscopy and cryo-electron microscopy can be applied to get detailed structural information on dynamic and heterogenous protein arrangements involved in virus assembly (in particular HIV-1) and membrane vesicle budding (COPI, COPII and Clathrin). Routes towards using cryo-electron tomography to solve structures in situ within complex environments.
