Comment[ArrayExpressAccession] E-GEOD-52702 MAGE-TAB Version 1.1 Public Release Date 2013-11-30 Investigation Title Interactive effects of arsenic and a high-fat diet on hepatic gene expression Comment[Submitted Name] Interactive effects of arsenic and a high-fat diet on hepatic gene expression Experiment Description Chronic exposure to inorganic arsenic (iAs) or a high-fat diet (HFD) can produce liver injury. However, the interactive molecular biological effects and mechanism of iAs and HFD are as of yet unclear. We used microarrays to detail the interactive effects of arsenic and a high-fat diet on hepatic gene expression. The C57BL/6 Mice fed low-fat diet (LFD) or HFD were exposed to 3 mg/L iAs or deionized water for 10 weeks. Then, hepatic RNA were extraction and hybridization on Affymetrix microarrays. Differentially expressed genes in LFD+As, HFD, and HFD+As groups compared to LFD group were identified, and interactive molecular biological effects and mechanism of iAs and HFD were discussed. GeneChips were washed and stained in the Affymetrix Fluidics Station 450 A total of forty mice were randomly assigned to four groups. Mice were fed a control LFD (13% fat in calories) or HFD (42% fat in calories) and given deionized water with or without sodium arsenite (3 mg iAs/L) for 10 weeks. Male C57Bl/6J mice were housed in stainless-steel cages under controlled conditions (25±3 °C, 50±5 % humidity and 12/12 h light/dark cycle) for 10 weeks Total RNA for each mouse was separately extracted using Trizol reagent protocol.10 mice per group were randomly divided into two sets (5 mice per set). Equal amounts of RNAs from 5 mice in the same set were pooled into one sample, and two samples per group were obtained. GeneChips were scanned using the GeneChip Scanner 3000.
