Experiment Description	IL-4 and IL-13 are cytokines involved in type 2 immune responses involved in atopic asthma and other diseases. They have a partially overlapping set of cognate receptors whose roles are incompletely understood. Here we explore the effects of blocking one or both subunits of the IL-13 alpha receptor on the action of these cytokines on IMR-90 lung fibroblast cells. IMR-90 lung fibroblast cells were cultured on matrigel and treated with various combinations of IL-4 (10 ng/ml), IL-13 (10ng/ml), 228 B/C (which blocks signaling from IL13RA1), and 11H4 (which blocks signaling from IL13RA1 and IL13RA2).

Protocol Description	The data were processed with Agilent Feature Extraction software, version 10.7.3.1, using default settings, including linear and Lowess normalization. ID_REF =  VALUE = Agilent log2 ratio, test channel (ch1)/reference channel (ch2).	Biotinylated cRNA were prepared according to the standard Agilent protocol.	cRNA was hybridized on Agilent-014850 Whole Human Genome Microarray 4x44K.	Cells were treated for 24 hours with TNFa (10 ng/ml). After this 24 hour TNFa pre-treatment, cells were treated with IL-4 (10 ng/ml), IL-13 (10ng/ml), 228 B/C (which blocks signaling from IL13RA1), and/or 11H4 (which blocks signaling from IL13RA1 and IL13RA2).	IMR-90 cells (ATCC, Manassas, VA; Catalog # CCL-186) were cultured in DMEM medium supplemented with 10% FBS (Sigma, St. Louis, MO; Catalog # F2442) and Penicillin/Streptomycin (Invitrogen, Carlsbad, CA; Catalog # 15140). Cells were plated on a bed of growth factor reduced Matrigel (GFR Matrigel, BD Biosciences, Bedford, MA; Catalog No. 354230).	RNeasy Mini Kit (Qiagen).	Stratagene Universal Human Reference RNA (catalog number 740000).	Chips were scanned using the Agilent Surescan Microarray Scanner.
