Experiment Description	To characterize the nature of the unfolded protein response in this Toxoplasma, we carried out microarray analyses to measure the changes in the transcriptome and in translational control during ER stress. Total RNA or RNA fractionated into monosomes or polysomes were purified from parasites treated with DMSO or tunicamycin for 1 hour.  RNA was processed for microarray analysis using a ToxoGeneChip.

ID_REF =  VALUE = MAS5 signal ABS_CALL = indicating whether the transcript was present (P), absent (A), or marginal (M) DETECTION P-VALUE = 	total RNA was labeled using the 3' IVT express kit from Affymetrix.	Affymetrix standard protocol, except hybridization for 17 hours.	Purified parasites were treated with 10 uM tunicamycin or DMSO for 1 hour	extracellular RH tachyzoites were purified from human fibroblast by filtration	Total RNA or RNA from sucrose fractions were ethanol precipitated and purified using Rneasy columns	Scanned using 3000-7G controlled by Affymetrix GCOS.
