Experiment Description	Gene expression analysis was conducted on lung RNA from C57BL/6J mice and compared to lung RNA from B6.129S2-Trp53tm1Ty/J mice (p53 NULL) to observe differences in baseline gene expression in mice with p53 deficiency. Lung RNA was isolated at baseline (saline airway exposure) in a total of 5 biological reps for both C57BL/6J (WT) and p53-deficient mice.

Protocol Description	Data was obtained using the Agilent Feature Extraction software (v9.5), using the 1-color defaults for all parameters. ID_REF =  VALUE = Processed Cy3 signal intensity (Agilent gProcessedSignal)	Starting with 500ng of total RNA, Cy3-labeled cRNA was produced according to the manufacturer’s protocol.	For each sample, 1.65ug of Cy3-labeled cRNAs were fragmented and hybridized for 17 hours in a rotating hybridization oven.	8-week-old male mice received aerosolized saline for 30 min prior to collection of lung for RNA processing.	Whole lung was collected and flash frozen in liquid nitrogen. Total lung RNA was isolated using the Qiagen RNeasy kit following the manufacturer's recommendations.	Slides were washed and then scanned with an Agilent Scanner.
